Mechanism of Action
Diclofenac inhibits the synthesis of prostaglandins by inhibiting the cox-1 and cox-2 enzyme and prevents the formation of PGG2 which the precursor for the synthesis of other prostaglandins.
Metaxalone causes central nervous system depression.
Both the drugs are absorbed orally.
Diclofenac is mainly bound to serum albumin.
Diclofenac is more than 99% bound to human serum proteins, primarily to albumin. Serum protein binding is constant over the concentration range (0.15-105 µg/mL) achieved with recommended doses. Although plasma protein binding and absolute bioavailability of metaxalone are not known, the apparent volume of distribution (V/F ~ 800 L) and lipophilicity (log P = 2.42) of metaxalone suggest that the drug is extensively distributed in the tissues.
Metaxalone is metabolized in the liver whereas diclofenac undergoes oxidation and conjugation.
Both the drugs are Excreted via urine
- Dark color urine
- Pale or yellow skin
- Stomach pain
- Contraindicated in renal impairment
